As a home based, client facing Business Development Manager, you will focus your efforts on the acquisition of borrowers through intermediaries (Commercial Finance Brokers etc). more specifically, you will develop an intermediary acquisition strategy; identify which intermediaries to target, in what order, and how and assist with campaign/event planning throughout the year to deliver on set strategy . you will acquire and build relationships with brokers both by telephone and face to face, field and respond to queries from brokers, educate intermediaries on maximising our client?s product and ultimately seek to increase the number of brokers and frequency of use. Long gone are the days when restaurant franchising was purely synonymous with fast food burger joints and pizza parlours. The industry has now branched out and restaurant franchises include not only themed food from countries all across the globe, but also internet cafes, coffee stalls, tea shops, sandwich bars and cookie outlets. Many involve several food and drink types, combining coffee, smoothies, juice and other drinks with muffins, bagels, baguettes and wraps. One of the newest restaurant franchising opportunities in the UK focuses on the health trend, with all menu items having some health benefit. Healthy eating venues emphasise their use of fresh, quality nutritionally balanced, high-flavour ingredients and pride themselves on their ?green? credentials, their aim being to run a responsible and sustainable business. Prices at restaurants and cafes specialising in healthy food tend to be higher than those at fast food outlets and are therefore usually found in affluent areas, or where consumers are more likely to be health or image conscious, such as health clubs, gyms, leisure centres and private clinics or hospitals. NHS hospitals have in the past rented out their catering spaces to fast food restaurant franchises, but these contracts are mostly due to end within the next two to three years. There is mounting pressure on the Government to take more action on obesity and health lobbyists have been campaigning for an end to fast food outlets in hospitals. It appears, therefore, that there will be a gap in the market and one which healthy eating restaurant franchises will be eager to fill. So it appears very likely that the health food restaurant franchising business is set for success in the UK.

Storm forces GOP to scrap first day of convention

TAMPA, Fla. (AP) ? Republican officials abruptly announced plans Saturday night to scrap the first day of their national convention, bowing to the threat of Tropical Storm Isaac as it bore down menacingly on Florida.

"The safety of those in Isaac's path is of the utmost importance," tweeted Mitt Romney, his formal nomination as presidential candidate pushed back by a minimum of 24 hours from Monday night to Tuesday.

The announcement was made as delegates and other convention-goers flocked to the Tampa Bay area by the planeload for what had been scripted as four days of political pageantry and speechmaking with a purpose ? to propel Romney into the fall campaign against President Barack Obama.

Officials said they hoped to begin laying out a revised schedule on Sunday.

Romney campaigned in battleground Ohio during the day, pledging to help female entrepreneurs and innovators who are eager to create small businesses and the jobs that go with them. It was an economy-themed countdown to a convention taking shape in a city already bristling with security ? and bracing for a possible hurricane.

"Women in this country are more likely to start businesses than men. Women need our help," said the Republican presidential challenger, eager to relegate recent controversy over abortion to the sidelines and make the nation's slow economic recovery the dominant issue of his convention week.

Reince Priebus, the Republican Party chairman, told reporters on an early evening conference call that no state delegations had changed their travel plans because of the storm. "Everyone is planning on being here and we hope we are up and running and expect all of our delegates to be here," he said.

Yet with rain and high winds in the forecast, and with the threat of a storm surge and possible flooding, convention organizers said they were making contingency plans to move delegates who have been booked into beachfront hotels to other locations if necessary.

North Dakota Republican Party Chairman Stan Stein said he needed instructions on preparing emergency kits for the 65 to 70 people in his state's contingent staying at a hotel 200 yards off the water in Treasure Island. "We do blizzards. We can handle them, but we're just going to go on the advice we're getting from hotel management on how to prepare for this," he said.

"Our first priority is ensuring the safety of delegates, alternates, guests, members of the media attending the Republican National Convention, and citizens of the Tampa Bay area," convention CEO Bill Harris said in an emailed announcement that followed private conversations involving Romney's campaign, Florida Gov. Rick Scott's office, security officials and others.

The announcement said that while the convention would officially be gaveled into session on Monday as scheduled, the day's events would be cancelled until Tuesday.

The announcement made the GOP convention the party's second in a row to be disrupted by weather. Four years ago, the delegates gathered in St. Paul, Minn., but Hurricane Gustav, slamming the Gulf Coast, led to a one-day postponement.

In that case, party officials rewrote their script to make President George W. Bush's speech into a video appearance, and to cancel plans for Vice President Dick Cheney to appear before the delegates. Both men were unpopular at the time.

Four years later, there was no immediate sign that Romney's forces would do anything other than squeeze two nights' of platform programming into one. Nor did it appear the postponement would cost them much in political terms, since the television networks had already announced they would not be carrying any of Monday's events live.

Despite the disruption, Priebus said, "we are optimistic that we will begin an exciting, robust convention that will nominate the Romney-Ryan ticket."

Plans had called for the convention to open Monday with quick ratification of a conservative platform expected, followed by Romney's nomination in a traditional roll call of the states timed for network evening news coverage.

Barring further postponements it will end Thursday with his prime-time acceptance speech, which aides hope will propel him into a successful fall campaign and, eventually, the White House.

The polls made the race a close one, narrow advantage to Obama, as two weeks of back-to-back conventions approached. Hundreds of millions of dollars have been spent on television ads, with hundreds of million more to come, almost all of it airing in a small group of battleground states expected to settle the election.

The list included Florida as well as North Carolina, where the Democratic National Convention will be held in one week's time.

Scott declared a state of emergency earlier in the day as the storm approached the Florida Keys, more than 400 miles from Tampa. Forecasters said it was on a track to head west of the convention city, but predicted strong winds and rain at a minimum on Monday as the delegates were to board buses for their first trip to the hall.

"We are a hospitality state. We know how to take care of people and we want to ensure their safety," Scott said.

Apart from weather concerns, a heavy security presence was already in evidence. Miles of fencing were designed to create a secure zone around a tract of land that included the convention hall, the hotel where Romney will stay and a nearby convention center where journalists and others worked.

Obama did his best to intrude on the Republican unity tableau.

In an interview with The Associated Press, he accused Romney of holding "extreme positions" on economic and social issues, while pledging a willingness on his own part to agree to "a whole range of compromise" with Republicans if he is re-elected.

He did not elaborate, but his pledge seemed designed to appeal to independents and other voters who say they are tired of seemingly perpetual campaign bickering and Washington gridlock.

Plans for Vice President Joe Biden to campaign in Florida were cancelled, also because of the threat posed by the storm.

But Romney said Obama's entire campaign rested on his ability to persuade people to ignore his record and listen instead to his rhetoric.

"It is not his words people have to listen to. It's his action and his record," he said in his appearance in Powell, Ohio. "And if they look at that, they'll take him out of the office and put people into the office who'll actually get America going again."

Romney's speech included an appeal to women made on economic grounds rather than on the basis of social issues like abortion, the sort of approach the Republican hopes will eat into Obama's polling advantage among female voters.

"I want to make sure that we help entrepreneurs and innovators. I want to speak to the women of America who have dreams, who begin businesses in their homes, who begin businesses out in the marketplace, who are working at various enterprises and companies," he said.

Romney envisioned an economic resurgence fueled by abundant energy, expanded trade and a skilled workforce. If that happens, "America is going to surprise the world. We're going to stand out as a shining city on a hill in part because of our extraordinary economy," he said to the cheers of an estimated 5,000 supporters.

Romney's determination to turn the attention to the economy follows two weeks of controversy over Medicare, courtesy of Obama's campaign, as well as abortion, the result of a comment by Rep. Todd Akin, the party's candidate in a Senate race in Missouri.

Romney joined an unsuccessful effort by party leaders to force Akin to quit his race after he said women who are raped rarely become pregnant, a view unsupported by medical evidence.

He also fought back hard in recent days in person and television advertising against Obama's allegations that he and running mate Ryan would remake Medicare in a way that would undermine the health of future seniors.

Power and Associates' Power Information Network? (PIN) and LMC Automotive.

Retail Light-Vehicle Sales
August new-vehicle retail sales are projected to come in at 1,066,200 units, which represents a seasonally adjusted annualized rate (SAAR) of 12.3 million units. The year-over-year growth rate in retail sales continues a double-digit trend for a fourth consecutive month. Retail transactions are the most accurate measurement of true underlying consumer demand for new vehicles.

"August continues this summer's trend of healthy growth in retail sales as dealers work to?sell down inventory in time to make room for 2013 models," said John Humphrey, senior vice president of global automotive operations at J.D. Power and Associates. "To date, automakers have been diligent in better balancing production with demand, which has been critical to the improved financial performance for many brands. Going forward, this discipline will be tested as demand looks to cool somewhat through the balance of the year."

While incentives are down slightly in August, compared with July ($106 less per vehicle, on average), there are deals driving some activity as the model-year sell down takes hold. Consumers are pushing aside the economic risks, as the need to replace their current vehicle is matched by availability of both inventory and credit.

Total Light-Vehicle Sales
Total light-vehicle sales remain stable, with the volume in August expected to come in at 1,285,300 units, a 16 percent increase from August 2011. Fleet represents only 17 percent of total light-vehicle sales, which is lower than the 21 percent year-to-date average.

Sales Outlook
LMC Automotive recently revised the outlook for total light-vehicle sales in the United States downward to 14.3 million units from 14.5 million units, with retail sales at 11.4 million units, down from 11.5 million units. Weaker economic growth and concerns with the European crisis are the driving factors for slower growth during the second half of the year. The industry is still expected to achieve the 15-million-unit level in 2013, but the outlook has been tempered from a projected 15.2 million units, as the risks in 2012 spill over into next year.

"The strength in August light-vehicle sales takes some of the pressure off expectations for the balance of the year, but a high level of risk lingers," said Jeff Schuster, senior vice president of forecasting at LMC Automotive. "We expect the current seesawing in auto sales to continue for the foreseeable future, but the overall picture in 2012 remains positive."

North American Production
North American light-vehicle production volume has increased by 23 percent through the first seven months this year, compared with the same period in 2011. The increase of nearly 1.7 million additional vehicles highlights the industry's recovery from the challenging production environment in 2011.

Production for Honda and Toyota in the first seven months of 2012 are up 79 percent and 65 percent, respectively, as recovery by both manufacturers takes hold. U.S. manufacturing growth is outpacing the rest of the North American region, with a 27 percent year-to-date increase as newer capacity drives the growth. Production in Mexico has increased 14 percent, with further growth expected as new key models ramp up. Canadian manufacturing has increased 20 percent year to date, but the level of future volume is at risk, as the domestic manufacturers and CAW begin labor negotiations.

Vehicle inventory in early August declined slightly to a 54-day supply, compared with 58 days in July. Car inventory remains at a below-normal level with a 47-day supply, down from 49 days in July. Truck inventory is at normal levels with a 61-day supply, down from 67 days in July.

"With the robust production activity outpacing the increases in demand, North American volume is approaching the 15.0-million-unit level for the first time since 2007," said Schuster. "However, given that inventory has normalized and growth in demand is expected to slow, LMC Automotive is holding the forecast for 2012 at 14.9 million units for the year."

Adze:
The adze is a vampiric being in Ewe folklore. No one knows what the adze look like in their real form, the legends do suggest however that if the adze is captured it will revert back to its true form. When in human form, the adze has the power to possess humans. The adze's influence would negatively affect the people who lived around their host. A person is suspected of being possessed in a variety of situations, including: women with brothers (especially if their brother's children fared better than their own), old people (if the young suddenly started dying and the old stayed alive) and the poor (if they envied the rich). The adze's effects are generally felt by the possessed victim's family or those the victim is jealous of.

The Analan:
The Analan are deformed spirits from the folklore of the Philippines. They have wings, and their fingers and toes point backwards. The Analan are said to take drops of menstrual blood, miscarried fetuses, afterbirth, or other reproductive waste and transform them into grotesque copies of human children and raise them as their own. According to legend the Analan live near springs in extremely fine houses, made of gold and other valuables.

Anti-April Fool:
April 2nd is Anti-April Fools Day and with it comes the evil mischief of the April Anti-Fool aka the nemesis of the April Fool who blesses us with humorous harmless pranks on April Fools Day (April 1st). Although the two are enemies they are separated temporally and confined to their respective days which for them lasts one whole year. Unlike the April Fool, the Anti-Fool can be very dangerous in nature and their so called pranks can be extremely dangerous and diabolical. The Anti-Fool has equivalent magical powers, including mind control, and enhanced superhuman abilities.

The Anti-Fool is also said to be able to mimic the faces of humans. Unlike the Fool whose powers are truly imbued within him, the Anti-Fool derives his powers from his scepter so taking it away will turn him into a normal evil human. If he is unable to recover the scepter within one minute then his face will permanently be molded into the image of his skull mask, rendering him ineffectual and unable to disgiuse himself by changing faces.

Leap Day Demon Goddess:
Chuang-Mu, the Chinese Goddess of Bedchambers, divorced her God husband Chuang-Kong some centuries ago, and decided to spice up her life by joining the demonic masses of the underworld. Generally Gods, and Demons steer clear of each other but every so often they come to an understanding. Chuang-Mu is now a rare Demon Goddess whose been on quite a destructive rampage for a great many centuries. Some of her most sinister handy works take place on the Leap Days of Leap Years.

On that fateful day she materializes as a beautiful young Asian woman who finds the most innocent guys possible. By the end of the day she seduces them with the ultimate intent on getting them to marry her in an unholy ceremony. Once joined with the Goddess in unholy matrimony they disappear just before the stroke of Midnight on March 1st. They are never seen again until the next leap day. Her victims speak of the hellish years spent with the sadistic demon in a plane of reality not of our own. She feeds of their souls to fuel her power, and gain favor with the Archdemonic hierarchy. During that day they have a chance to escape her evil clutches while she?s distracted searching for a new soul to corrupt.

In medieval legend, a cambion is a half-human offspring of a demon and a human. Caliban, in The Tempest by William Shakespeare, is a cambion; Merlin from the Arthurian legend is also a cambion. Most often depicted as the offspring of an incubus and a human woman. At birth, the infant has no pulse and no breath. This continues until the child is about seven years old, where it becomes increasingly difficult to differentiate one from a human. A cambion is usually devilishly cunning and angelically beautiful, able to persuade even the most strong-hearted individual to do his or her bidding. The method of the creation of a cambion is protracted. A succubus will have sex with a human male and so acquire a sample of his sperm. This she will then pass on to an incubus. The incubus will, in his turn, transfer the sperm to a human female and thus impregnate her.

Churel:
A churel is a female ghost (witch like) out of Hindu folklore. The spirit is said to be of a low-caste woman who died either in childbirth. More than anything, sustainability makes more business sense ? even to the financial bottom line.

Some CEOs, such as Paul Polman of Unilever, our 2011 Grand Prix Winner, clearly believe that sustainability is not only the right thing, but the smart thing for long term business stability and success. Polman has stated that he wants Unilever?s ambitious and laudable Sustainable Living Plan to act as a positive example for the business case for sustainability. Unilever are making good progress towards their targets; they plan to source 100% of agricultural raw materials sustainably by 2020, and have already increased the amount from 14% in 2010 to 24% last year, so it is no surprise that Unilever?s share price has steadily increased following adoption of the Sustainable Living Plan.

A more positive outlook is provided by Timetric?s Global CEO Sustainability Outlook Survey 2012. Along with being specifically focused on obtaining senior executives? opinions on the meaning, implementation, measurement and management of sustainability, the survey also links sustainability to profitability, thus connecting two business imperatives.. The survey found that 43% of overall industry suppliers expect to see an increase in profitability over the next 12 months due to implementation of sustainability, suggesting that in some cases, simply switching to more sustainable practices produced a financial return. Respondents were found to be influenced by cost, efficient operations, competitive advantage and client demand, so the decision for companies to become more sustainable will depend on changes having a positive impact on these factors.

The imperative that arises out of these surveys and discussions is to find the best way to align responsible, sustainable business with enhanced operations and financial returns. While it may be enough for companies to focus on the tried and tested actions such as waste reduction, recycling, more efficient water and energy use, and other traditional CSR actions, at the board level, a more radical approach is needed than simply adhering to the triple bottom line. Business leaders and CEOs need strong, visionary leadership in order to convince not only their employees but their shareholders that making moves towards sustainable business is sensible in terms of the long-term health and resilience of the company. In order to achieve this, they must believe it is not merely sensible, but right.

Forest razing by ancient Maya worsened droughts, says study

ScienceDaily (Aug. 21, 2012) ? For six centuries, the ancient Maya flourished, with more than a hundred city-states scattered across what is now southern Mexico and northern Central America. Then, in A.D. 695, the collapse of several cities in present day Guatemala marked the start of the Classic Maya's slow decline. Prolonged drought is thought to have played a role, but a study published this week in the journal Geophysical Research Letters adds a new twist: The Maya may have made the droughts worse by clearing away forests for cities and crops, making a naturally drying climate drier.

"We're not saying deforestation explains the entire drought, but it does explain a substantial portion of the overall drying that is thought to have occurred," said the study's lead author Benjamin Cook, a climate modeler at Columbia University's Lamont-Doherty Earth Observatory and the NASA Goddard Institute for Space Studies.

More than 19 million people were scattered across the Maya empire at its height, between A.D. 250 and A.D. 900. Using population records and other data, the study authors reconstructed the progressive loss of rainforest across their territory as the civilization grew. The researchers ran computer simulations to see how lands newly dominated by crops would have affected climate. In the heavily logged Yucatan peninsula, they found that rainfall would have declined by as much as 15 percent while in other Maya lands, such as southern Mexico, it would have fallen by 5 percent. Overall, the researchers attributed 60 percent of the drying estimated at the time of the Maya's peak to deforestation.

As crops like corn replace a forest's dark canopy, more sunlight bounces back into space, said Cook. With the ground absorbing less energy from the sun, less water evaporates from the surface, releasing less moisture into the air to form rain-making clouds. "You basically slow things down -- the ability to form clouds and precipitation," he said.

The idea that the Maya changed the climate by clearing away jungle, partly causing their demise, was popularized by historian Jared Diamond in his 2005 book Collapse. In the first study to test the hypothesis, climate modeler Robert Oglesby and his colleagues ran a computer simulation of what total deforestation of Maya lands would do to climate. Their results, published in 2010 in the Journal of Geophysical Research, showed that wet season rainfall could fall 15 to 30 percent if all Maya lands were completely cleared of trees. Oglesby, who was not involved in the Cook study, said that Cook's estimate of a 5 to 15 percent reduction in rainfall, though lower than his own, makes sense since Cook's simulation used a realistic tree-clearing scenario.

Archeologists attribute a variety of factors to the collapse of the Classic Maya, whose ancestors are still living today in parts of Mexico, Guatemala, Belize, El Salvador and Honduras. In addition to a drying climate in several regions, the city-states struggled with overpopulation, changing trade routes, war and peasant revolts.

The Maya cleared the forests to grow corn and other crops, but they also needed the trees for cooking large amounts of lime plaster used in constructing their elaborate cities. Thomas Sever, an archeologist at the University of Alabama, Huntsville, and a co-author of the 2010 deforestation study, said that it would have taken 20 trees to produce a single square meter of cityscape. "When you look at these cities and see all the lime and lime plaster, you understand why they needed to cut down the trees to keep their society going," he said.

The Maya also lacked the technology to tap the groundwater several hundred feet beneath them. Their reservoirs and canals were able to store and distribute water when rain plentiful, but when the rain failed, they had nowhere to turn. "By the time of the collapse, every square mile of soil had been turned over," said Sever.

Scientists know from studying climate records held in cave formations and lake sediments that the Maya suffered through a series of droughts yet they continue to debate their severity. In a paper earlier this year in Science, researchers Mart?n Medina-Elizalde and Eelco Rohling of Mexico's Yucatan Center for Scientific Research found that annual rainfall may have fallen as little as 25 percent during the Maya's decline, from about A.D. 800 to A.D. 950. Most of the reduction in rainfall, however, may have occurred during the summer growing season when rain would have been most needed for cultivation and replenishing freshwater storage systems, they added.

Today, many of the Maya's abandoned cities are overgrown with jungle, especially on the Yucatan peninsula. Satellite images, however, show that deforestation is happening rapidly elsewhere, including in other regions the Maya once occupied. The study may offer a warning about the consequences: "There's a tremendous amount of change going on in Guatemala," said Oglesby. "They may be that much more vulnerable to a severe drought."

Investing in Micro-Enterprise is a WISE Economic Growth Strategy ...

Last week the Associated Press reported an increase in the number of microloans disbursed in the United States since the economy began to falter in 2008. These microloans, sometimes as small as $50.00, are sourced by nonprofit organizations that often provide small business training for low-income microentrepreneurs, most of whom are women.

Federal economic policy is out of synch with these trends. The Small Business Administration (SBA)?s Program for Investment in Micro-Entrepreneurs (PRIME) offers technical assistance and support to nonprofit microfinance organizations. But the program, which is tiny to begin with, has experienced budget cuts and was nearly eliminated in fiscal year 2012. The current fiscal year 2013 budget makes the program increasingly vulnerable; if not eliminated, PRIME may experience additional cuts.

In fact, both Congress and the President?s job growth agendas have largely ignored microentreprise. While President Obama?s latest small business focus may help increase access to capital for some, these policies do not accomplish enough for low-income microentrepreneurs who have great potential but lack strong credit or business training.

Given that microenterprise can improve the lives of low-income women and their families, as well as boost the economy, PRIME and other pro-microenterprise initiatives ought to take a greater role in the federal government?s economic growth strategy.

Cultivating microenterprise has a proven return on investment in the United States, as many organizations have demonstrated: Women?s Initiative for Self Employment (WISE), Erase Poverty, and more famous entities like Grameen Bank, and Accion, are just a few of the groups that provide low-income entrepreneurs with microloans, as well as training and support in creating viable businesses.

WISE, founded in 1988, is exclusively focused on supporting low-income women entrepreneurs in the San Francisco Bay Area. WISE?s program offers a range of supports to low-income women, including training on building a business plan and the nuts and bolts of launching a business, as well as microloans. To date, WISE graduates have launched 1,600 small businesses that pay an average of $16.45 per hour, double the minimum wage in California.

Adriana Lahl is a 2010 graduate of WISE?s training program. Lahl founded Sal de Vida, a company that produces gourmet salts, spreads, and rice mixes. Lahl now belongs to the highly competitive program with La Cocina, another San Francisco microenterprise NGO that supports low-income food entrepreneurs.

Lahl has a 6-year-old daughter who is on MediCal, and her husband is unemployed. She is hopeful her company can become steady source of income for her family over the long term.

?WISE helped me build my business plan and provided me with a $3,000 loan that I am now paying back,? Lahl said. ?It was important to me to begin building relationships and credit for my company.?

WISE Interim CEO Nicole Levine tells me that her organization?s resources have been constrained in recent years (and they expect their PRIME funding to decline), even as demand for WISE?s program is increasing among low-income women.?

Programs like WISE offer a level of support that low-income women simply can?t get from traditional lenders, like commercial banks that cater to people who already have resources, or at least good credit.

And a ?small? loan from traditional lenders is still at least tens of thousands of dollars. It is not possible for many low-income women to qualify for a loan of that size. Unlike traditional loans, a microloan can be a much smaller loan at low interest.

Even a $250.00, $1,000 or $3,000 loan, along with support resources, can change the lives of a woman and her family. As the Associated Press story pointed out, ?A 2008 survey by the Aspen Institute found the average household income of families participating in a microenterprise program rose 20 percent to $36,000.?

If we take further advantage of the economic potential of microenterprises, many, many Americans could benefit. According to two major microenterprise advocacy organizations, the Association for Enterprise Opportunity and the California Association for Micro Finance Opportunity (CAMEO), if one in three microenterprises hired just one more person, the US would have zero unemployment. As it stands, the very smallest enterprises that employ fewer than 5 people account for 17 percent of the GDP and employ 31 million people.

Many federal programs are experiencing cuts these days, and PRIME may continue to feel the squeeze in 2013. But the program should not be eliminated. PRIME, along with other support for microenterprise, should become a more central part of the President and Congress?s long-term economic growth strategy--so that groups like WISE can cultivate more businesswomen like Adriana Lahl.

A compound already sitting on the shelves of biomedical laboratories and emergency room supply closets seems to interrupt the formation of neurodegenerative protein clumps found in Huntington?s disease, according to a preliminary animal?study?published August 7 in the?Journal of Neuroscience.

This versatile agent, called methylene blue, gets a mention in medical literature as?early?as 1897 and was used to treat, at one time or another, ailments ranging from?malaria?to cyanide?poisoning. The U.S. Food and Drug Administration has never formally approved it as a therapy for any illnesses. But that fact hasn?t stopped biomedical researchers from tinkering with the agent?s apparent ability to?improve?cognitive function. And although the new paper out today relies on a Huntington?s disease model in flies and mice, scientists are hopeful. "Because of existing knowledge of methylene blue and the fact that it?s not harmful to humans, I would hope that progress toward clinical trials could go relatively quickly," says?Leslie Thompson, a neurobiologist at University of California?Irvine and lead author on the new study.

Huntington?s?disease?occurs when the C-A-G sequence of DNA base pairs repeat too often on the?HTT?gene, resulting in an abnormally long version of the huntingtin protein, that therefore folds incorrectly and forms clumps in the brain. The illness usually begins to affect people in their 30s and 40s, causing movement problems and early death. No drug is currently available to stop the disease from progressing.

For their experiment, researchers fed methylene blue mixed with food for a week to?Drosophila?flies engineered to carry a mutated copy of the?HTT?gene. An ?examination of the flies? brains showed that protein clumps had been reduced by 87 percent compared with a control group. Meanwhile, mice designed to carry the mutated gene were given methylene blue and underwent several tests to assess mobility. At two months of age, the treated mice showed abnormal clasping of their hind claws only 20 percent of the time in a reflex test, whereas their untreated counterparts clasped at a 60 percent rate. (Less clasping meant healthier mice.) While the number of mice used was not sufficient to provide statistically significant results and the difference in the test quickly dropped off at nine weeks of age, Thompson still views the data as hopeful, because even a delay in Huntington?s symptoms would be very helpful. Thompson quickly points out that more research is needed. "The early steps of aggregation [protein clumping] are getting altered in a test tube, in flies and in mice?and that?s significant," she says, and speculates that methylene blue may possibly prevent the mutant?Htt?protein from sticking to itself.

What these findings mean for the approximately 25,000 to 30,000 individuals with Huntington?s disease in the U.S. right now is that a small molecule agent like methylene blue that has been grandfathered into approved use as a diagnostic tool in humans can be studied further as possible treatment for the neurodegenerative illness. "This study shows promise pre-clinically and follow-up studies are needed in a more representative mouse model that expresses the full-length Huntingtin protein and allows more study of the disease?s neurogenerative properties," says?Albert La Spada, a neurogeneticist at the University of California?San Diego. Thompson underscores this point as well: "Methylene blue would absolutely require further testing in mouse models and would need safety and efficacy trial before it could be used for humans."
